Precision analysis for standard deviation measurements of immobile single fluorescent molecule images.

چکیده
Standard deviation measurements of intensity profiles of stationary single fluorescent molecules are useful for studying axial localization, molecular orientation, and a fluorescence imaging system's spatial resolution. Here we report on the analysis of the precision of standard deviation measurements of intensity profiles of single fluorescent molecules imaged using an EMCCD camera. We have developed an analytical expression for the standard deviation measurement error of a single image which is a function of the total number of detected photons, the background photon noise, and the camera pixel size. The theoretical results agree well with the experimental, simulation, and numerical integration results. Using this expression, we show that single-molecule standard deviation measurements offer nanometer precision for a large range of experimental parameters.

Simultaneous, accurate measurement of the 3D position and orientation of single molecules.
Recently, single molecule-based superresolution fluorescence microscopy has surpassed the diffraction limit to improve resolution to the order of 20 nm or better. These methods typically use image fitting that assumes an isotropic emission pattern from the single emitters as well as control of the emitter concentration. However, anisotropic single-molecule emission patterns arise from the trans...
متن کاملSingle-Molecule Fluorescence Microscopy Reveals Local Diffusion Coefficients in the Pore Network of an Individual Catalyst Particle
We used single-molecule fluorescence microscopy to study self-diffusion of a feedstock-like probe molecule with nanometer accuracy in the macropores of a micrometer-sized, real-life fluid catalytic cracking (FCC) particle. Movies of single fluorescent molecules allowed their movement through the pore network to be reconstructed. The observed tracks were classified into three different states by...

Background & Aim: Measuring the length of kidneys is an important issue in diagnosis and treatment of renal diseases. In various diseases, kidneys become larger or smaller than normal. Ultrasonography is a suitable method for measuring renal length because it is accurate, noninvasive, available and with low cost. Nowadays, multislice spiral CT scanners are widely used for evaluation of rena...
متن کاملA three-dimensional digital image correlation technique for strain measurements in microstructures.
A three-dimensional digital image correlation technique is presented for strain measurements in open-cell structures such as trabecular bone. The technique uses high-resolution computed tomography images for displacement measurements in the solid structure. In order to determine the local strain-state within single trabeculae, a tetrahedronization method is used to fill the solid structure with...
متن کاملPrecision of distance determination using 3D to 2D projections: The error of migration measurement using X-ray images
The goal of this study has been to objectively and reliably estimate the precision of measuring 2D migration of hip prostheses. This is the distance change over time between the implant and the bone observable in X-ray images. To reach this goal, a generally valid scheme for determining the standard deviation of distance measurements in 3D to 2D projections has been worked out. The scheme was a...
